A handle is the difference between carrying a camera and operating one. It moves the weight onto a proper grip and puts the rig at eye level without a tripod.
Mounting points on the handle itself carry a microphone, a small light or a monitor, so the accessory travels with your hand rather than fighting it.
A NATO clamp comes off in one movement when the camera has to go on a gimbal; a threaded mount is more rigid and slower to change.
